RT–PCR analysis of NIH-3T3 cells transfected with retrofitted BAC DNA. Cells were transfected with DNA from retrofitted BAC clone 261M17, which contains p53 genomic sequences (P53 BAC) or BAC clone 67L19 (Control), cultured for 4 weeks in the presence of G418 and pooled for analysis. RT–PCR using primers specific for murinep53 (right) was positive for an expected 194-bp product in both transfections. RT–PCR using primers specific for humanp53 (left) was positive for an expected 199-bp product only in cells that were transfected with P53 BAC DNA. All lanes were negative when reverse transcriptase was not added.
