Endogenous gene disruption through CPP-mediated delivery of Cas9 protein and guide RNA in several cell lines. Cell lines such as HEK293T (human embryonic kidney cells), HeLa (human cervical cancer cell line), and NCCIT (human embryonal carcinoma cell line) cells (A), and primary cells such as human dermal fibroblasts (HDF) and human embryonic stem cells (B), were treated with Cas9-m9R and sgRNA:9R once per day for 3 d. The gene disruption efficiencies were determined using the T7E1 assay 3 d after the first treatment. The arrow indicates the expected position of DNA bands cleaved by T7E1.
