Cover Marathon runners represent the dynamics of the transcription process, influenced by DNA sequence elements. In this issue, single-cell time-lapse microscopy experiments were used to shed light on the ways in which two types of promoter elements—nucleosome disfavoring sequences and transcription factor binding sites—influence transcription dynamics. Promoter dynamics dictate the transcriptional noise across a cell population, as illustrated by the variability between the runners. (Cover illustration by Maya Dadiani.
