Cover In this issue, the pattern of de novo cancer mutations and evolutionary mutations in human mitochondrial genomes are compared. It is observed that recurrent combinations of cancer mutations preferentially coincide with ancient haplotypes, with evidence for similar selective constraints, suggesting a functional potential for these mutations.
