Interspecies sequence homology at the 5′ end of the bli-4gene. Sequence alignment of regions I–VII within the respective 5′ ends of the Ce–bli-4 (Ce) and Cb–bli-4 (Cb) genes are available online at www.genome.org. For clarity, a conservedBglII site is shown in boldface lettering. A schematic representation of constructs is used to examine the transcriptional regulation of a bli-4–lacZ reporter gene. The plasmid pCeh320 contains 5219 bp of the Ce–bli-4 5′ end, in which a series of deletion clones have been constructed. The relative level of β-galactosidase expression exhibited by transgenic strains that carry each plasmid is shown as high (+++), low (+), variable (+/−), or not observed (−).
