FFPEcap-seq overview. During enzymatic pretreatment, RNA is treated with T4 Polynucleotide Kinase (PNK) to phosphorylate 5′ hydroxyls and Terminator Nuclease to degrade uncapped RNAs. Sequence tags are added to the 5′ end of the cDNA during the reverse transcription reaction using template switching. cDNA is then amplified, and additional sequences are added during PCR amplification. (OH) Hydroxyl; (P) phosphate; (Gr) a guanine ribonucleotide; (N) an equal mix of nucleotides.
