Overview of the BS-tagging assay. Standard transposases are used but with a 5mC fill-in. The bisulfite treatment changes the original transposon sequence and prevents it from being amplified, but the fill-in complement with 5mC remains amplifiable. One custom oligonucleotide is needed for this assay.
