Our experimental system for measuring promoter activity conservation across species. Native RP promoters from nine different yeast species (including S. cerevisiae) were extracted and inserted into a fixed genomic location on chromosome 15 of S. cerevisiae, immediately upstream of a YFP reporter gene. YFP levels were measured in vivo, enabling promoter activity differences of as little as 10% to be significantly detected (see Zeevi et al. 2011).
