Six-nucleotide triads form the basis of canonical triplex formation. The third nucleotide forms Hoogsteen or reverse Hoogsteen hydrogen bonds with the purine of the duplex, resulting in parallel and anti-parallel orientation of the third strand to the purine tract of the duplex, respectively.
