SL-PCR amplification of 5′ segments of trans-spliced mRNAs. Starting from a capped (m*Gppp) and polyadenylated (AAAAAAAAAAn) trans-spliced mRNA (top line) with the 16-nt SL sequence at its 5′ terminus, the diagram illustrates the basis of AN-linked random-hexamer-primed reverse transcription, SL-primed second strand cDNA synthesis, and PCR amplification with SL and AN primers.
