Promoter-reporter assay. Predicted miRNA promoters or control sequences were cloned into pGL3 enhancer vector containing SV40 enhancer, but no promoter. Plasmids were transfected into Jurkat cells together with Renilla luciferase control. Luciferase activity was measured after two days.
