Schematic of sample-specific barcode PCR. Sample-specific DNA barcodes are incorporated into the primers that are used for the final universal PCR. The 5′ end of the universal primer (white) is tailed with the sequences for the 454 FLX machine (gray) and sample-specific DNA sequences (black). When sequencing from either 454A or 454B, the first few bases indicate from which sample the read originated.
