DNaseI HS mapping in skin epidermis. Nuclei were obtained from the epidermis of Klf4-/- and wild-type newborn mice and treated with increasing amounts of DNaseI. The purified DNA was used for Southern blot analysis. The HSs detected by probe P324 on PstI-restricted DNaseI-treated DNA and probe P370 on SspI-restricted DNaseI-treated DNA in epidermis are the same as in the SP-1 cell line. Quantifying the intensity of the HSs bands in DNA digested to comparative levels indicates that the HSs are more prevalent in Klf4-/- than in the wild-type DNA.
