Schematic for GST preparation. In this method, DNA is first fragmented with a rare cutter such as NotI or a more frequent cutter such as BamHI. Specific complementary biotinylated linkers are ligated to the free ends, and the DNA is then digested withNlaIII. All subsequent steps in the protocol are identical.
