The general principle behind different pyrosequencing reaction systems. A polymerase catalyzes incorporation of nucleotide(s) into a nucleic acid chain. As a result of the incorporation, a pyrophosphate (PPi) molecule(s) is released and subsequently converted to ATP, by ATP sulfurylase. Light is produced in the luciferase reaction during which a luciferin molecule is oxidized.
