Downstream application of cloned ORFeome and promoterome clones. MultiSite Gateway Recombinational cloning technology allows transfer of attL1-ORF-attL2 and attL4-promoter-attR1 Entry clones either independently, to attR1-ccdB-attR2 and attR4-ccdB-attL1 destination vectors respectively, or together into a single attR4-ccdB-attR2 destination vector, while preserving the reading frame and orientation. The availability of a cloned C. elegans “promoterome” and the corresponding “ORFeome” should facilitate a vast number of in vivo assays in the worm, such as ectopic expression, subcellular localization, tissue specific RNAi, and tissue specific rescue experiments.
